(1). 抗原或抗体的固相化及抗原或抗体的酶标记。
(2). 结合在固相载体表面的抗原或抗体仍保持其免疫学活性。
(3). 酶标记的抗原或抗体既保留其免疫学活性,又保留酶的活性。
(4). 受检标本与固相载体表面的抗原或抗体起反应。再加入酶标记的抗原或抗体,也通过反应而结合在固相载体上。
(5). 此时固相上的酶量与标本中受检物质的量呈一定的比例。
(6). 加入酶反应的底物后,底物被酶催化成为有色产物,产物的量与标本中受检物质的量直接相关,故可根据呈色的深浅进行定性或定量分析。测定方法具有很高的敏感度(pg-ng/ml水平),并且重复性好。以免疫学反应为基础,将抗原、抗体的特异性反应与酶对底物的高效催化作用相结合起来的一种敏感性很高的试验技术。

Phosphorylated upon DNA damage, probably by ATM or ATR. Phosphorylated on Ser-15 upon ultraviolet irradiation; which is enhanced by interaction with BANP. Phosphorylated by NUAK1 at Ser-15 and Ser-392; was initially thought to be mediated by STK11/LKB1 but it was later shown that it is indirect and that STK11/LKB1-dependent phosphorylation is probably mediated by downstream NUAK1 (PubMed:21317932). It is unclear whether AMP directly mediates phosphorylation at Ser-15. Phosphorylated on Thr-18 by isoform 1 and isoform 2 of VRK2. Phosphorylation on Thr-18 by isoform 2 of VRK2 results in a reduction in ubiquitination by MDM2 and an increase in acetylation by EP300. Stabilized by CDK5-mediated phosphorylation in response to genotoxic and oxidative stresses at Ser-15, Ser-33 and Ser-46, leading to accumulation of p53/TP53, particularly in the nucleus, thus inducing the transactivation of p53/TP53 target genes. Phosphorylated at Ser-315 and Ser-392 by CDK2 in response to DNA-damage.
Dephosphorylated by PP2A-PPP2R5C holoenzyme at Thr-55. SV40 small T antigen inhibits the dephosphorylation by the AC form of PP2A.
May be O-glycosylated in the C-terminal basic region. Studied in EB-1 cell line.
Ubiquitinated by MDM2 and SYVN1, which leads to proteasomal degradation.
