背景介绍:Killer cell immunoglobulin-like receptors (KIRs) are transmembrane glycoproteins expressed by natural killer cells and subsets of T cells. The KIR genes are polymorphic and highly homologous and they are found in a cluster on chromosome 19q13.4 within the 1 Mb leukocyte receptor complex (LRC). The gene content of the KIR gene cluster varies among haplotypes, although several "framework" genes are found in all haplotypes (KIR3DL3, KIR3DP1, KIR3DL4, KIR3DL2). The KIR proteins are classified by the number of extracellular immunoglobulin domains (2D or 3D) and by whether they have a long (L) or short (S) cytoplasmic domain. KIR proteins with the long cytoplasmic domain transduce inhibitory signals upon ligand binding via an immune tyrosine-based inhibitory motif (ITIM), while KIR proteins with the short cytoplasmic domain lack the ITIM motif and instead associate with the TYRO protein tyrosine kinase binding protein to transduce activating signals. The ligands for several KIR proteins are subsets of HLA class I molecules; thus, KIR proteins are thought to play an important role in regulation of the immune response.

1.阳性细胞染色分布有三种类型:细胞浆、细胞核、细胞膜表面。大部分抗原见于细胞浆,可见于整个胞浆或部分胞浆
2.阳性细胞分布可分为灶型和弥漫性
3.由于细胞内含抗原量不同,所以染色强度不一。如果细胞之间染色强度相同,常提示其反应为非特异性
4.阳性细胞染色定位于单个细胞,且与阴性细胞相互交杂分布;而非特异性染色常不限于单个细胞,而是累及一片细胞。
5.切片边缘、刀痕或褶皱区域,坏死或挤压的细胞区,胶原结缔组织等,常表现为相同的阳性染色强度,不能用于判断阳性。
